- 利用SuperScript? II RNase H- RT的先進性能，提高第一鏈cDNA產量，增加cDNA長度
- 在一個反應中，利用至多1 μg的總RNA或poly(A)+ RNA制備特定的cDNA
- 提供更完善的第一鏈cDNA純化步驟，獲得適合加尾的cDNA
- 在與PCR相容的緩沖液中，以優化的反應條件使用重組TdT，為PCR提供高效的引物退火位點
