Transcription factor Sox2 is a member of sox(sry-related HMG box-containing)gene family. Sox2 functions in diverse important developmental processes such as early embryogenesis, neural differentiation and lens development.Sox2 is critically involved in the signaling pathway for maintaining self-renewal and pluripotency of embryonic stem (ES) cells. Sox2 contains an HMG domain in its N-terminal region that recognizes and bends the same DNA sites as SRY and has a transactivation domain at its C terminus. Sox2 and its closely related genes (Sox1 and Sox3) are classified as subfamily group B genes. Sox2 regulates a number of target genes by binding to respective regulatory regions and it has been shown to work in concert with other transcription factors including Oct4 as well as Nanog to support stem cell potency and self-renewal.
            


            ·Description

            Recombinant Human Sox2 produced in E. coli is a single, non-glycosylated, polypeptide chain containing 317 amino acids and having a molecular mass of 34.3kDa. The Recombinant Human Sox2 is produced as C-terminal poly-arginine fusion protien.
            


            ·Quality Control
            
Purity: Greater than 98.0% as determined by: 

            (a) Analysis by RP-HPLC. 

            (b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. 

            Amino-Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was foundto be Met-Trp-Asn-Met-Met.
            
Endotoxin:Less than 0.1ng/μg (1.0EU/μg) of rHuSox2 as determined by LAL test.

·Formulation

            SteriLyophilized from a 0.2um filtered protein solution in PBS.
            
·Reconstitution

            It is recommended to reconstitute the lyophilized rHuSox2 in sterile PBS not less than 100μg/ml, which can then be further diluted to other solutions.
            


            ·Storage

            yophilized rHuSox2 although stable at room temperature for 3 weeks, should be stored desiccated below-18°C. Upon reconstitution recombinant human Sox2 should be stored at 4°C between 2-7 days and aliquots ofreconstituted samples with 0.1% HSA or BSA are stable up to 3 months below -18°C for future use.
