美國BIOLOG公司獨(dú)家研發(fā)的產(chǎn)品 ECO板,特別為微生物群落分析和生態(tài)研究設(shè)計,Eco板上包含國際土壤微生物學(xué)家精選的用于微生物群落分析最常用的31種碳源,是國際上微生物生態(tài)功能多樣性研究的最典型和最權(quán)威方法之一。
每塊96孔ECO板上有3組平行,每組31種碳源,3個陰性對照,加入同一樣品后,可以獲得3組平行數(shù)據(jù)。微生物群落對這31種碳源的特征性利用稱作該微生物群落的代謝指紋圖譜。從單個的微平板上的指紋圖譜就可以獲得大量的代謝信息。可用于純種及混合菌群(如土壤樣品)微生物功能多樣性研究。
31種碳源可分為六大類,具體如下:

樣品簡單處理后(過濾或離心),取上清液加到ECO板上培養(yǎng)多至5天時間,每隔一定時間用酶標(biāo)儀在590nm和750nm下測定OD值,OD值的大小反應(yīng)樣品微生物利用某一碳源底物的能力,將OD值導(dǎo)出后進(jìn)行分析,如AWCD值(平均吸光度)、豐富度、單孔的動力學(xué)分析、不同類碳源的比較等,并可用聚類軟件對不同樣品進(jìn)行聚類分析。
值得一提的是ECO板不僅測定可培養(yǎng)微生物,還可測定部分不可培養(yǎng)微生物,只要微生物能利用碳源進(jìn)行代謝(并不增殖),ECO板上顯色體系均可檢測出來。
最常見的應(yīng)用是利用ECO板分析環(huán)境因素、根系、葉面、肥料、農(nóng)藥對土壤微生物菌群的生態(tài)功能多樣性的影響,比較不同樣品間或同一樣品不同時間的功能多樣性差異,比較環(huán)境修復(fù)前后微生物群落的功能多樣性差異,等等。
