Brain-derived neurotrophic factor (BDNF) is a member of the neurotrophin family. Along with other structurally related neurotrophic factors NGF, NT-3 and NT-4, BDNF binds with high affinity to the TrkB kinase receptor. It also binds with the LNGFR (for low-affinity nerve growth factor receptor, also known as p75). BDNF promotes the survival, growth and differentiation of neurons.It serves as a major regulator of synaptic transmission and plasticity at adult synapses in many regions of the cns.BDNF expression is altered in neurodegenerative disorders such as Parkinson's and Alzheimer's disease.


Description
Recombinant Human Brain-derived neurotrophic factor produced in E. coli is a non-glycosylated polypeptide chain containing 119 amino acids.Bioactive BDNF is a non-covalently linked homodimer and having a molecular mass of 27kDa.


Quality Control
Biological activity: The ED50 of rHuBDNF as determined by cell proliferation assay using the TrkB transfected BaF3 mouse pro-B cell line was 1-6ng/mL. 
Purity: Greater than 98.0% as determined by: 
(a) Analysis by RP-HPLC. 
(b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. 
Amino-Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-His-Ser-Asp-Pro . 
Endotoxin: Less than 0.1ng/μg (1EU/μg) of hBDNF as determined by LAL test. 

Formulation
Sterile and Lyophilized aliquots with a 0.2um filtered protein solution containing 20mM PB pH8.0, 400mM NaCl.

Reconstitution
It is recommended to reconstitute the lyophilized rHuBDNF in sterile 18MΩ-cm H2O not less than 100μg/ml, which can then be further diluted to working solutions.

Storage 
Lyophilized rHuBDNF although stable at room temperature for 3 weeks, should be stored desiccated below -18°C, which is stable for up to twelve months from date of receipt. Upon reconstitution rHuBDNF should be stored at 4°C between 2-7 days and aliquots of reconstituted samples are stable up to 3 months below -18°C for future use.
