Background:
The accumulation of the amyloid-beta (AB)
plaque in the cerebral cortex is a critical event in
the pathogenesis of Alzheimer's disease. Ab
peptide is generated by proteolytic cleavage of
the
and
sites by two proteases. APP is first
cleaved by
-secretase, producing a soluble
derivative of the protein and a membrane
anchored 99-amino acid carboxy-terminal
fragment (C99). The C99 fragment serves as a
substrate for
secretase to generate the 5 kDa
amyloid-
peptide, which is deposited in the
brains of all patients with Alzheimer's disease.
The long-sought
-secretase was recently
identified by several groups independently and
designated
-site APP cleaving enzyme (BACE)
and aspartyl protease 2 (Asp2). BACE/Asp2 is
a novel transmembrane aspartic protease and
co-localizes with APP.
Specificity:
This antibody can be used for detection of BACE.
Human brain tissue lysate can be used as a
positive control with a band at approximately 70
kDa.

Human and mouse, others not tested.

Immunogen:
This rabbit polyclonal antibody was raised
against a peptide corresponding to amino acids
485 to 501 of human BACE. The peptide
sequence differs from those of mouse and rat
BACE by one amino acid.

The IgG has been purified by the use of ion
exchange chromatography. The concentration
is 100 μg IgG in 200 μL of PBS containing
0.02% sodium azide.
Storage and Stability:
Stable for one year when stored at 4°C.
Applications and Suggested Dilutions:

Western blotting: 0.5-1 μg/mL
