皮膚上皮層是一層保護(hù)機(jī)體免受外界環(huán)境損傷的重要屏障。該層主要為角質(zhì)細(xì)胞類型,包含了大約85%活的表皮細(xì)胞。角質(zhì)層是復(fù)扁平上皮細(xì)胞。角質(zhì)細(xì)胞因含有大量的角蛋白而得名,在表皮基底層,角質(zhì)細(xì)胞分化、增殖,隨著角質(zhì)細(xì)胞產(chǎn)生角蛋白并上移至表皮表面后,發(fā)生程序性死亡。角蛋白細(xì)胞增殖、分化及凋亡是復(fù)雜且嚴(yán)格控制的過(guò)程。角蛋白細(xì)胞可產(chǎn)生很多細(xì)胞因子和生長(zhǎng)因子,包括IL- 1, IL-3, IL-6, IL-8, 集落刺激因子, TNF-alpha, TGF-alpha and beta, 成纖維細(xì)胞生長(zhǎng)因子, ?雙調(diào)蛋白和PDGF。除了保護(hù)功能之外,角蛋白細(xì)胞在組織穩(wěn)態(tài)、傷口恢復(fù)、癌癥以及皮膚基因治療上有著重要的作用。人角蛋白細(xì)胞能表達(dá)粘附分子和細(xì)胞因子,這說(shuō)明了它能參與皮膚免疫和動(dòng)態(tài)平衡。
??????? <2100>人表皮角質(zhì)細(xì)胞(HEK-n)-新生和<2110>人表皮角質(zhì)細(xì)胞(HEK-a)分別提取于新生兒與的表皮組織,于原代凍存。每管含有細(xì)胞數(shù)>5×105 cells/ml,此細(xì)胞通過(guò)Cytokeratine-8, -18 and -19免疫熒光染色驗(yàn)證,經(jīng)測(cè)試不含有HIV-1、HBV、HCV、支原體、細(xì)菌、酵母和真菌。細(xì)胞可以達(dá)到15倍增。

The epithelial layer of the skin provides an essential function as a protective barrier against insult from the outside environment. The major cell type in this layer is keratinocytes which consists around 85% of living epidermal cells. Keratinocytes are the cells of stratified squamous epithelia. It is so named because its most abundant protein is keratin. Keratinocyte divide and are thus generated in the basal layer of the epidermis; this division is followed by a programmed death as they produce keratin and move up to the surface of the epidermis. Keratinocyte proliferation, differentiation, and apoptosis are complex and carefully choreographed processes [1]. Besides the protective function, keratinocytes also play a central role in tissue homeostasis, wound healing, cancers, and skin-based gene-therapy. Human keratinocytes express adhesion molecules and cytokines indicating their participation in skin innate immunity and homeostasis in vivo [2, 3].
HEK-a from ScienCell Research Laboratories are isolated from adult human epidermal tissue. HEK-a are cryopreserved on primary culture and delivered frozen. Each vial contains >5 x 10^5 cells in 1 ml volume. HEK-a are characterized by immunofluorescent method with antibodies to cytokeratine-18 and -19. HEK-a are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HEK-a are guaranteed to further expand for 15 population doublings in the condition provided by ScienCell Research Laboratories.

Directly and immediately transfer cells from dry ice to liquid nitrogen upon receiving and keep the cells in liquid nitrogen until cell culture needed for experiments.
