Macrophage colony-stimulating factor 1, also  known as M-CSF,CSF-1, Lanimostim, is a single-pass membrane protein  which is disulfide-linked as a homodimer or heterodimer. M-CSF is a  cytokine and acts as a primary regulator of macrophage survival,  proliferation and differentiation. It is also essential for the survival  and proliferation of osteoclast progenitors. M-CSF plays important  roles in immunological defenses, bone metabolism, lipoproteins  clearance, fertility and pregnancy. Recombinant M-CSF containing N  terminal portion of M-CSF is sufficient for interaction with the M-CSF  receptor. Human M-CSF is active in the mouse, but mouse M-CSF is  reported to be species specific.




·Description

Recombinant Human M-CSF produced by  transiently transfected human cell consists of sequence  (Accession#P09603)(Glu33-Arg255) of human M-CSF fused with a  polyhistidine tag at the C-terminus.




·Quality Control


Biological activity:The ED50 of rHuM-CSF is 0.5-1.5 ng/mL,as measured in a cell proliferation assay using M-NFS-60 mouse myeloid cells.


Purity: Greater than 90.0% as determined by: 

(a) Analysis by RP-HPLC. 

(b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. 

Amino-Acid Sequence: Predicted N terminal amino acid is Glu33.


Endotoxin:Less than 0.1ng/μg (1EU/μg) of rHuM-CSF.

·Formulation

Sterile and Lyophilized from a 0.2 μm filtered solution in PBS.


·Reconstitution

It is recommended to reconstitute the  lyophilized rHuM-CSF in sterile water not less than 100μg/ml, which can  then be further diluted to other aqueous solutions.


·Storage 

Lyophilized rHuM-CSF although stable at room  temperature for 3 weeks, should be stored desiccated below -18°C. Upon  reconstitution rHuM-CSF should be stored at 4°C between 2-7 days and  aliquots of reconstituted samples containing 0.1% BSA are stable up to 3  months below -18°C for future use.
