Background:
DNA polymerase ? has an 8 kDa amino-terminal
ssDNA binding domain with deoxyribose
phosphodiesterase activity and a 31 kDa
carboxy-terminal domain with
nucleotidyltransferase activity. Mammalian DNA
polymerase ? is constitutively expressed in
cultured cells, but levels can be up-regulated by
treating cells with DNA-alkylating agents (eg.
methyl methanesulfonate). DNA polymerase ?
fills single nucleotide gaps in DNA produced by
the base excision repair pathway of mammalian
cells.
Specificity:
Monoclonal antibody recognizes the 39 kDa
DNA Polymerase ?. Shows no effect on the
polymerase activity of ?-pol.

Applications and Suggested Dilutions:
Functional Activity: Effect on Polymerase
Activity, Non-inhibitory.
Western blot: Use 1-2 μg/mL for 2 hrs. at RT.
Immunoprecipitation (Denatured verified) Use
protein G; Ab at 2 μg/mg protein lysate.
The optimal dilution for a specific application
should be determined by the researcher.
