| cDNA第一鏈反轉(zhuǎn)錄試劑盒采用穩(wěn)定高效的反轉(zhuǎn)錄預(yù)混體系5×Golden RT MasterMix進(jìn)行RNA的反轉(zhuǎn)錄反應(yīng)，使用時(shí)只需加入模板、引物和RNase Free H2O 即可，大大簡(jiǎn)化了操作過程、提高了效率、減少了操作過程中的人為誤差。MasterMix中添加了HL dsDNase（熱敏雙鏈特異性核酸酶），該雙鏈特異性核酸酶（又名gDNA Remover）可在反轉(zhuǎn)錄的過程中，直接降解去除RNA中殘留的基因組DNA污染(可去除100ng基因組DNA)，從而在定量PCR的檢測(cè)中無需考慮基因組污染的干擾，設(shè)計(jì)的定量PCR引物也無需進(jìn)行橫跨外顯子。該試劑盒尤其適用于定量PCR檢測(cè)。 該預(yù)混體系包含點(diǎn)突變致RNase H活性缺失的Golden MLV Reverse Transcriptase反轉(zhuǎn)錄酶、dNTP、反應(yīng)Buffer、gDNA去除試劑和RNase Inhibitor。該試劑盒采用的反轉(zhuǎn)錄酶去除了RNase H活性，從而避免反轉(zhuǎn)錄過程中降解RNA。同時(shí)經(jīng)過突變文庫篩選，使得其熱穩(wěn)定性更強(qiáng)，可耐受55℃高溫反應(yīng)。相比于低溫條件下反轉(zhuǎn)錄反應(yīng)，采用高溫反轉(zhuǎn)錄可顯著打開RNA二級(jí)結(jié)構(gòu)，從而提高復(fù)雜RNA模板的擴(kuò)增性能、提高反轉(zhuǎn)錄cDNA的長(zhǎng)度和產(chǎn)量，從而提高后續(xù)檢測(cè)的靈敏度。合成的第一鏈cDNA可廣泛用于2nd Strand的合成、雜交、PCR擴(kuò)增、Real-Time PCR反應(yīng)等。 |
