嵌合抗體(Chimeric Antibody)是一種通過基因工程技術(shù)將來自不同物種的抗體基因片段融合,以創(chuàng)建具有所需特性(如高親和力、低免疫原性)的新抗體分子。

如人鼠嵌合抗體,是由鼠源性抗體的V 區(qū)基因與人抗體的C 區(qū)基因拼接為嵌合基因,然后插入載體,轉(zhuǎn)染骨髓瘤組織表達(dá)的抗體分子。通常是將小鼠可變區(qū)外顯子(包含抗原結(jié)合位點(diǎn))與人類恒定區(qū)外顯子(負(fù)責(zé)抗體的效應(yīng)功能)融合,構(gòu)建嵌合免疫球蛋白基因。最終生產(chǎn)一種能夠逃脫人體免疫系統(tǒng)監(jiān)視,減少對人體免疫系統(tǒng)刺激性的新抗體,同時保留小鼠單克隆抗體的特異性。
