斑馬魚作為腸道損傷模型的應用已得到充分證實,葡聚糖硫酸鈉(Dextran Sulfate Sodium Salt,DSS)誘導的斑馬魚IBD模型模擬了哺乳動物IBD表型的不同特征,包括腸道中性粒細胞炎癥、過多粘液生成、腸淋巴管生成增加以及前炎性細胞因子上調。

實驗材料
DSS(Yeasen 60316ES,MW:36000~50000)
實驗步驟
1. 將斑馬魚胚胎培養在含甲基藍的E3胚胎培養基中,28.5 ℃,培養至1 dpf;
2. 1dpf 后使用不含亞甲基藍的E3胚胎培養基,培養至3 dpf;
3. 用E3培養基配制濃度為10% DSS的儲存液;
4. 用培養基稀釋DSS至最大非致死劑量(DSS濃度參考:0.5%);
5. 用0.5%的DSS處理斑馬魚,從3 dpf處理到6 dpf,觀察指標[1]。
結果分析
造模成功指標:中性粒細胞數量增多、酸性粘液蛋白增多、炎性因子表達量上升。
1.嗜中性粒細胞數量:中性粒細胞數量增多,并且向腸和上皮中移動
2.酸性粘液蛋白:石蠟病理切片結合AB-PAS染色法驗證,DSS浸泡后斑馬魚腸道中酸性粘液蛋白增多
3.炎性因子表達量:DSS處理后斑馬魚腸道中炎性因子表達量上升[1,2]

DSS處理組相較于對照組,酸性粘蛋白染色和粘液分泌增加,斑馬魚外周血中的嗜中性粒細胞數量增多并且向腸和上皮中移動,炎性因子表達量顯著上調,腸道出現炎癥反應。
