Descriptions：

?? Brdu，中文全名5-溴脫氧尿嘧啶核苷，為胸腺嘧啶的衍生物，可代替胸腺嘧啶在DNA合成期（S期），活體注射或細胞培養加入，而后利用抗Brdu單克隆抗體，ICC染色，顯示增殖細胞。同時結合其它細胞標記物，雙重染色，可判斷增殖細胞的種類，增殖速度，對研究細胞動力學有重要意義。

APPLICATIONS:

BrdU is selectively incorporated into cell DNA at the S phase of cell cycle. The use of BrdU as a thymidine analog has made possible the identification of DNA synthesis in suspensions of cells, cell smears and tissue sections. The incorporation of BrdU into DNA in place of thymidine is discussed by A.L. Givan.5 BrdU at 0.16 to 500 mg/mL of cell culture media produced inhibition of growth of KD cells (rabbit kidney cells). Effective inhibition at concentrations greater than 1.0 mg/mL was observed.6 It is incorporated, in vivo,by injecting 10-100 mg/kg at 10 mg/mL in saline intraperitoneally.7 It is also incorporated into bone marrow cells in culture at a final concentration of 10 mM at 37EC for one hour. For incorporation to occur, the BrdU must be phosphorylated in the cell by thymidine kinase.8 FITC-conjugated second antibodies can be used with antibodies specific for BrdU (Sigma product B2531)

which will make "new" DNA fluoresce green; denatured DNA can be stained with propidium iodide and will fluoresce red.
