?鈣黃綠素 AM 是一種膜滲透性熒光酯酶底物,通常用于檢測細胞活性和質膜完整性。作為一種近中性的底物,鈣黃綠素 AM 可以自由、無創地擴散穿過質膜并滲透到活細胞的胞質溶膠中。一旦進入細胞內,代謝活躍細胞中的胞質酯酶會迅速將非熒光鈣黃綠素?AM?底物水解成鈣黃綠素產物,并由具有完整質膜的細胞保留。由于死細胞缺乏酯酶活性,因此僅標記活細胞。在藍光激發下,鈣黃綠素發出強烈的綠色熒光,與細胞酯酶的活性和樣品中存在的活細胞數量成正比。與其他活細胞標記試劑(如?BCECF AM?或羧基熒光素二乙酸酯)相比,鈣黃綠素?AM?熒光更亮、光穩定性更好,低細胞毒性有利于細胞追蹤、細胞凋亡和細胞毒性的研究。
