小鼠模型是結腸炎造模常見模型,自從1985年首次采用葡聚糖硫酸鈉(dextran sulphate sodium, DSS)制備出倉鼠潰瘍性結腸炎模型后,已有大量數據證明DSS結腸炎模型的病因、臨床癥狀、病理改變及治療應答均與人類潰瘍性結腸炎(ulcerative colitis,UC)相似。
現行研究中常用的UC建模類型大約分為:自發模型、誘導模型(化學藥物誘導:TNBS三硝基苯磺酸,DSS葡聚糖硫酸鈉等、免疫學方法誘導、細胞移植誘導)以及基因修飾模型等。
DSS造模優勢
葡聚糖硫酸鈉鹽(Dextran Sulfate Sodium Salt, DSS)是一種聚陰離子衍生物,其誘導結腸炎模型的機制雖仍未十分明確,但通常認為與巨噬細胞功能失調、腸道菌群失調、DSS對結腸上皮的毒性作用、細胞因子在DSS結腸炎模型的發病中起重要作用等機制有關。
對比多種UC建模類型,DSS造模具有很大優勢:
1、癥狀與人類UC高度相似:可用于研究結腸炎發生機制和藥效研究;2、成模率高:自由飲用DSS水溶液,簡單易行、重復性強;3、可構建多種結腸炎模型:急性結腸炎、慢性結腸炎,還可以聯合AOM構建結腸炎相關性癌癥(CAC)模型;4、適用于多種屬動物造模:小鼠、大鼠、斑馬魚、豬、果蠅等;5、安全性高:DSS可被自然生態系統降解,對環境安全。

DSS建模實驗步驟

圖1. 小鼠結腸炎造模方案 DSS濃度:急性期模型通常為3-5%,慢性期模型通常為1-3%;可根據實驗需求及動物類型摸索濃度。該濃度為質量體積比,使用飲用水配置后,0.22µM濾膜過濾除菌。
數據記錄:每隔24h記錄小鼠體重、糞便粘稠度、糞便潛血等指標。

急性DSS結腸炎模型
Day1:稱重并標記各組C57BL/6小鼠。待造模小鼠飲用3-5% DSS水溶液,未處理組小鼠飲用正常水。Day3:給予待造模小鼠更換新鮮3-5%DSS飲用水。Day5:給予待造模小鼠更換新鮮3-5%DSS飲用水。Day8:給予待造模小鼠更換新鮮不含DSS的飲用水。

慢性DSS結腸炎模型
Day1:稱重并標記各組C57BL/6小鼠。待造模小鼠飲用1-3% DSS水溶液,未處理組小鼠飲用正常水。Day3:給予待造模小鼠更換新鮮1-3%DSS飲用水。Day5:給予待造模小鼠更換新鮮1-3%DSS飲用水。Day8:給予待造模小鼠更換新鮮不含DSS的飲用水。Day22-Day26:重復Day1-Day5操作。Day29:給予待造模小鼠更換新鮮不含DSS的飲用水。Day43-Day47:重復Day1-Day5操作。Day50:給予待造模小鼠更換新鮮不含DSS的飲用水。
