自噬是一種由應激誘導的保護性機制。在基礎條件下活性較低;當細胞面臨營養耗竭、化學或環境脅迫等特定不利條件時,會啟動該機制,通過溶酶體介導大量降解細胞內容物。自噬活性增強在癌癥、神經退行性疾病、心血管疾病和糖尿病病理中的作用已得到廣泛認可和普遍研究。
CYTO-ID® Autophagy detection kit利用一種新型染料,選擇性標記積累的自噬泡,從而檢測自噬泡并監測自噬流,適用于溶酶體功能受抑制的活細胞。這種可被488nm激發的綠色染料經過優化,通過鑒定其可調控的功能基團,實現了對溶酶體的極微染色,同時在結合前自噬體、自噬體和自噬溶酶體時呈現明亮的熒光。

PROTEOSTAT® Aggresome detection kit(貨號:ENZ-51035)
Aggresome是由聚集的錯誤折疊蛋白質形成的包涵體,通常在泛素-蛋白酶體蛋白質降解機制失效時形成。Aggresome是細胞為應對應激而產生的一種細胞保護機制,通過隔離錯誤折疊的蛋白質,最終使其通過自噬途徑被清除。此外,蛋白質Aggresome的形成也是多種人類疾病的標志,如阿爾茨海默病、帕金森病、肌萎縮側索硬化癥或酒精性肝病等。
PROTEOSTAT® Aggresome detection kit所含的PROTEOSTAT® 檢測試劑是一種分子染料。 在溶液中,染料分子沿單一中心鍵的自由旋轉會抑制熒光。PROTEOSTAT®染料能特異性嵌入到四級蛋白質結構特有的交叉β-折疊骨架中——這種結構常見于錯誤折疊和聚集的蛋白質——并產生強烈的熒光。該試劑盒提供了一種快速、特異且定量的方法來標記細胞內的Aggresome,無需對蛋白質進行人工突變。
