三磷酸腺苷結(jié)合盒轉(zhuǎn)運(yùn)體(ATP-binding cassette transporters, ABC)是一組跨膜蛋白,與ATP結(jié)合在胞膜和內(nèi)質(zhì)網(wǎng),過氧化物酶體,線粒體等細(xì)胞器膜上,介導(dǎo)氨基酸、脂質(zhì)、脂多糖、無機(jī)離子、多肽-糖類、各種藥物等多種分子的耗能轉(zhuǎn)運(yùn)。目前已知的人類ABC基因包括A、B、C、D、E、F、G7個(gè)家族,共48個(gè)成員,ABC轉(zhuǎn)運(yùn)體的變異可以相應(yīng)引起多種疾病,比如囊性纖維病,腦白質(zhì)腎上腺萎縮癥等。近年來,人們發(fā)現(xiàn)一些ABC家族成員與脂質(zhì)代謝有關(guān),并參與膽固醇結(jié)石等疾病的發(fā)生、發(fā)展過程。
ABCA1基因位于9q31,1,約149kb,其編碼一種含有2261個(gè)氨基酸的跨膜蛋白,主要參與膽固醇逆轉(zhuǎn)運(yùn),即HDL將外周組織過多的膽固醇轉(zhuǎn)運(yùn)到肝臟的代謝過程。ABCA1將細(xì)胞內(nèi)膽固醇和磷脂轉(zhuǎn)運(yùn)至細(xì)胞表面的HDL前體aooA1,該蛋白負(fù)責(zé)膽固醇逆轉(zhuǎn)運(yùn)的第一步也是主要的限速步驟,因此在膽固醇和血漿脂代謝中起著重要的作用。ABCG5和ABCG8是近年被發(fā)現(xiàn)的參與植物固醇和膽固醇吸收的基因,與β-谷固醇血癥有關(guān),這對(duì)基因定位在染色體2p21上,僅相距374個(gè)氨基酸,兩者都含有13個(gè)外顯子和12個(gè)內(nèi)含子,分別編碼sterolin-1和sterolin-2蛋白,它們?cè)诟闻K的肝細(xì)胞、膽管細(xì)胞、小腸上皮細(xì)胞以及膽囊上皮細(xì)胞中表達(dá)。
